  1. 1. Multiple-Emitter Super-resolution Imaging using the Alternating Descent Conditional Gradient Method

    University essay from Lunds universitet/Beräkningsbiologi och biologisk fysik - Genomgår omorganisation; Lunds universitet/Institutionen för astronomi och teoretisk fysik - Genomgår omorganisation

    Author : Dolev Illouz; [2023]
    Keywords : super-resolution; alternating descent conditional gradient method; ADCG; multiple-emitter localization; nanoscale imaging; single molecule tracking; Physics and Astronomy;

    Abstract : This thesis examines the state-of-the-art 2D super-resolution technique alternating descent conditional gradient (ADCG) method's ability to accurately localize fluorophores in diffraction-limited single molecule images (SMI) and analyze the impact of pre-processing and post-processing modules on ADCG's fluorophore localization. A synthetic dataset obtained from the 2013 Grand Challenge localization microscopy and a temporally linked dataset obtained from an unpublished set of Optical DNA mapping experiments performed by Jonathan Jeffet at the NanoBioPhotonix Lab at Tel-Aviv University were initially segmented to extract their noise parameters. 2. 2. Detection of Sclerotinia sclerotiorum in oilseed rape using Oxford Nanopore sequencing and qPCR

    University essay from Högskolan i Skövde/Institutionen för biovetenskap

    Author : Ana Guadalupe Herrera Hernandez; [2023]
    Keywords : ;

    Abstract : Sclerotinia sclerotiorum is a notorious phytopathogenic fungus and is the causal agent of the disease Sclerotinia stem rot (SSR) of rapeseed (Brassica napus). SSR is one of the main diseases affecting the yield and oil quality of rapeseed crops worldwide. READ MORE

  3. 3. Developing Automated Cell Segmentation Models Intended for MERFISH Analysis of the Cardiac Tissue by Deploying Supervised Machine Learning Algorithms

    University essay from KTH/Kemi

    Author : Julia Rune; [2023]
    Keywords : Cell Segmentation; Cellpose; Supervised Machine Learning; MERFISH; Heart Failure; Cellsegmentering; Cellpose; Övervakad Maskininlärning; MERFISH; Hjärtsvikt;

    Abstract : Följande studie behandlar utvecklandet av automatiserade cellsegmenteringsmodeller med avsikt att identifiera gränser mellan celler i hjärtvävnad. Syftet är att möjliggöra analys av data genererad från multiplexed error-robust in situ hybridization (MERFISH). READ MORE

  4. 4. Single-molecule Imaging of the Cell Division Ring in Escherichia coli Using the ALFA-tag

    University essay from KTH/Tillämpad fysik

    Author : Emma Westlund; [2023]
    Keywords : ALFA-tag; Escherichia coli; Divisome protein FtsZ; mEos3.2; Nanobodies; Nanotags; PALM; Plasmid transformation; sfGFP; Single-molecule Imaging; ALFA-tag; Escherichia coli; Divisomprotein FtsZ; mEos3.2; Nanokroppar; Nanotaggar; PALM; Plasmidtransformation; sfGFP; Enmolekyl-mikroskopi;

    Abstract : The use of super-resolution (SR) microscopy is an important tool for understanding the inside mechanisms of bacterial cells. However, for SR imaging, the labelling of the proteins of interest is a great challenge as flourescent proteins (FPs) are often too big to be directly fused to the target protein and traditional immunolabelling with antibodies creates too long separation between the fluorophore and the target protein. 5. 5. Study of Automated Step Detection Methods and Dwell Time Analysis of Single-Molecule Data : Case of Study: The Turnover of Single Fluorescent ATP on The Active Site of Myosin

    University essay from Linnéuniversitetet/Institutionen för fysik och elektroteknik (IFE)

    Author : Lathifa Nur Ramdhania; [2022]
    Keywords : Single-molecule; step detection; dwell time analysis;

    Abstract : Single-molecule methods were developed with the objective to obtain kinetic information of biomolecular processes from a single biomolecule. This objective can be achieved by analyzing the binding events in recordings (“trajectories”) of single molecules (e.g., using fluorescence); the so-called dwell times. READ MORE
